Characterization, Antioxidant and ?-Glucosidase Inhibitory Activity of Collagen Hydrolysate from Lamuru (Caranx ignobilis) Fishbone

The Lamuru fish (Caranx ignobilis) is mostly found in tropical waters of the indo-pacific region, namely Indonesia. It believed to contain collagen and this study aims isolate from its bone determine collagen’s antioxidant ?-glucosidase inhibitory activity. In our study, was extracted using acetic acid which hydrolyzed by collagenase enzyme Clostridium histolyticum at a temperature 37 °C, pH 7.0. During hydrolysis, degree hydrolysis (DH) calculated hydrolysates were characterized SDS -PAGE, UV-Visible spectroscopy FT-IR spectroscopy. After characterization, hydrolisate lamuru ( CHL) analyzed for properties result shows that higher percentage obtained, 31.17%, 120 min hydrolysis. CHL characterization -PAGE showed molecular weight ranging 35,000-180,000 Daltons identified as type I. UV-Vis analysis provided maximum absorbance wavelength 233 nm. At same time, presence amides I, II, III, confirms formation triple helix. For bioactivity assay, DPPH radical reduction activity reaching 51.45±1.24% (IC50 485.9 µg/mL). ferric power (FRAP value) significant Fe3+ Fe2+ with value 711.27 µM/g. inhibition IC50 determined be 574 µg/mL. Based on inhibition, peptide enables use therapeutic development variety disorders caused oxidative stress, such diabetes mellitus.
